A Modified Elution Method for Determining the Presence of Fetal Red Blood cells

Abstract
Coulter channelyser has been used to recognize the difference in size between maternal and fetal blood cells and to rapidly confirm the fetal origin of the aspirated blood sample. However, such a channelyser is not available in every center and
hospital
We wish to develop to a 6 and 7 min modified Kleihauer-betke technique which can be used for rapidly determining the presence of fetal red blood cells.
In 6-minutes methods, we used 80% Ethanol for fixation for 2.5min, elution solutation(0.4% ferric chloride and 0.5% hematoxylin), and staining solution(0.1% erythrosin). More accurate differentiation of fetal red blood cells were found in
7-minuted
method, suing ethanol for fixation for 3min, elution solution for 13 seconds and staining solution for 3 min, respectively.
